Teoricamente, il succo di pompelmo può aumentare le concentrazioni plasmatiche di loratadina, come avviene per altri farmaci che sono substrati del CYP450 3A4 via metabolica. Il meccanismo proposto è l'inibizione del CYP450 3A4-mediata metabolismo di primo passaggio nella parete intestinale da parte di alcuni composti presenti nei pompelmi. Il significato clinico di questo potenziale interazione è sconosciuto. Segnalate interazioni con potenti del CYP450 3A4 inibitori, quali claritromicina, eritromicina e ketoconazolo hanno prodotto un incremento sostanziale nella zona sotto il plasma curva concentrazione-tempo (AUC) della loratadina e del suo metabolita attivo, descarboethoxyloratadine, senza cambiamenti associati in generale il profilo di sicurezza del farmaco.
